Properties of DNA Fragments that are Associated with Valine Resistance to Acetohydroxy Acid Synthase Activity in Escherichia coli K-12

Escherichia coli (E. coli) strain K-12 in minimal salt medium is inhibited by L-valine. This growth sensitivity is largely attributed to the sensitivity of acetohydroxy acid synthase (AHAS) to feedback inhibition by valine. A cloned DNA fragment from E. coli that possesses valine resistant AHAS activity thus can support growth of valine-supplemented minimal medium. In this study, the cloned fragment was transformed into different strains of E. coli and monitored to see if these transformants acquired the abiltiy to grow on valine minimal media plates.
